The latest Paediatric Strategy Forum<\/strong>, organised by ACCELERATE<\/strong> in collaboration with the European Medicines Agency (EMA)<\/strong> and with participation from the FDA<\/strong>, has brought to light the current limitations in the development of cyclin-dependent kinase (CDK) inhibitors<\/strong> in paediatric oncology. Patricia Blanc<\/strong>, founder of the Imagine for Margo<\/strong> association, contributed to the publication of the Forum\u2019s conclusions in a landmark scientific article published in the European Journal of Cancer<\/em><\/a>.<\/p>\n

Many clinical trials, yet limited meaningful outcomes<\/h3>\n

The Forum critically assessed the many studies conducted in recent years on CDK4\/6 inhibitors<\/strong> (palbociclib, ribociclib, abemaciclib) in children with cancers such as neuroblastoma<\/strong>, gliomas<\/strong>, or sarcomas<\/strong>. While these molecules are widely used in adults\u2014especially for breast cancer\u2014the results in paediatrics remain modest, with mostly cytostatic effects<\/strong> (tumour growth slowdown) and very few actual tumour regressions<\/strong> observed.<\/p>\n

A strong message from patients: \u201cIt\u2019s time to change the paradigm\u201d<\/h3>\n

Patricia Blanc and other patient advocacy representatives emphasised the lack of coordination<\/strong> between trials, the redundancy of study designs<\/strong>, and the limited value gained<\/strong> given the number of children enrolled.<\/p>\n

\u201cExposing children to repeated, low-efficacy treatments without gaining real knowledge is a missed opportunity,\u201d stated Patricia Blanc during the Forum. \u201cWe need to prioritise, collaborate, and most importantly, learn from the past to avoid making the same mistakes again.\u201d<\/p>\n<\/blockquote>\n
